Title of PhD thesis: Production and manipulation of a recombinant bispecific antibody which binds simultaneously to leptin receptor and one of the T lymphocytes CD markers in order to modulate the immune response.

Summary of PhD thesis:

Leptin is one of the most important adipose derived hormones with a pivotal role in regulating energy homeostasis and metabolism, including appetite and starvation. Besides this weight control, leptin also plays an important role in the regulation of immune responses. In healthy individual with hypoleptinaemia leads to immunodeficiency and increased frequency of infection and hyperlptinaemia influences T cells that in turn lead to pro-inflammatory cytokines increase and probably involves in the onset and progressing of autoimmune diseases. Leptin has pro-inflammatory effects on T cell populations, shifting the T helper balance towards T helper-I phenotype, by inducing release of pro-inflammatory cytokines. Prevention of leptin binding to its receptor by an anti-leptin receptor monoclonal antibody (mAb) as a leptin antagonism suppresses some inflammatory disorders and it may offer a reasonable therapeutic approach. There is developmental expression of leptin receptor in different tissues particularly in the ventromedial nucleus of the hypothalamus; known as the "appetite center" and using any kind of leptin receptor blocker with antagonistic activity may result in side effects such as induction of considerable weight gain.

The aim of this study was production of leptin receptor inhibitor which affects specifically on T cells.

In this study a recombinant bispecific antibody was designed, using two kinds of hybridoma which produce monoclonal antibody against leptin receptor and T-helper lymphocyte CD4. ScFvs of anti-leptin receptor and anti-CD4 were produced separately, using phage display technique. Then they were both cloned in a phagemid vector as one protein, and finally bispecific antibody was expressed in E.coli. Western blot, ELISA and flow cytometry demonstrated specificity of the bispecific antibody. Also Mixed Lymphocyte Reaction (MLR) test was performed to evaluate the effect of bispacific antibody on interferon gamma (IFN-γ) as an important inflammatory cytokine marker of T lymphocytes. Bispecific antibody was able to reduce production of IFN-γ in MLR.
